While the first one is on your quest board, you need to find a secret note to activate the 100 floor quest- which can only be found after you've reached floor 25. Blow stuff up with bombs. Tip: start using bombs in addition to your pickaxe. Craft them with ores you don't need until you can buy some from the Dwarf. One time it was level 7, another level 40 something. But it usually has at least 10 Dino's.
